  • Overview An internally quenched fluorogenic peptide substrate (Mca/Anb as fluorophore/quencher) that is preferentially cleaved by cathepsin G between Gnf and Ser with high efficiency and selectivity ( k cat = 0.524 s -1 ; K M = 2.08 µ M; k cat / K M = 252, 000 M -1 s -1 ) as well as sensitivity (≤ 70 pM Cath G; [subsrate] = 3.3 µ M). Human neutrophil elastase and proteinase 3 are found to cleave the peptide between Val and Thr with a much slower kinetics (no detectable cleavage in 1 h) and lower efficiency (10% and 23% cleavage, respectively, after 24 h). Excitation max: 325 nm; emission max: 400 nm.
