  • Overview A highly sensitive fluorescence resonance energy transfer (FRET) peptide substrate, based on the EVNLDAEF sequence derived from the β-secretase site of the Swedish mutation of Amyloid Precursor Protein (APP). Hydrolysis of the substrate (Leu-Asp cleavage) by β-secretase results in fluorescence enhancement. k cat /K m values (min -1 /µ M) at 37° C, pH 4.5, for pro-memapsin 2 and its activated species Leu 28p -memapsin 2 are 0.044 and 0.040, respectively.
