Glucocorticoid receptor (UniProt P04150; also known as GR, Nuclear receptor subfamily 3 group C member 1) is encoded by the NR3C1 (also known as GCCR, GCR, GCRST, GRL) gene (Gene ID 2908) in human. Glucocorticoid receptor is widely e x pressed in almost all cell types and regulates genes that control development, metabolism, and immune response upon binding glucocorticoids. Hormone-bound GR translocates from the cytoplasm to the nucleus where it regulates gene transcriptions by binding to glucocorticoid response elements (GREs) or by interacting with various transcription factors. Alternatively, hormone-bound GR can represses gene transcriptions by blocking the nuclear translocation of transcription factors. BDNF-stimulated TrkB activation is shown to induce GR phosphorylation at Ser155 and Ser287 in primary rat cortical neurons (equivalent to human GR Ser134 and Ser267), resulting in enhanced GR transcription activity. GR is composed of an N-terminal modulating domain (a.a. 1-420), a DNA-binding domain (a.a. 421-486), and a C-terminal ligand-binding domain (a.a. 528-777).
