Our research has shown that high level protein e x pression can be achieved consistently when two processes, cell e x pansion and protein e x pression, are kept separate.
The Dual Media Set, different from commonly used protein e x pression procedures using Luria-Bertani (LB) medium or other specially prepared medium, contains two specially formulated media: E x pansion Broth (EB) and Overe x pression Broth (OB). For e x pansion, E. coli cells are grown in EB which keeps the production of recombinant protein repressed. To initiate high level protein e x pression, OB is simply added to the culture. By using the Dual Media Set, protein overe x pression can be reliably controlled for many recombinant proteins. In some circumstances, when the e x pressed protein is either to x ic or insoluble, overe x pression may be counter-productive. In such cases, protein production can be kept at a minimum by adding the inducer IPTG (for lac-based promoters) to cells growing in EB.
